Inclusion technology is widely used at present. Inclusion technology refers to the technology that one molecule is embedded in the hole structure of another molecule to form an inclusion complex. This inclusion complex is composed of the addition of main molecules and guest molecules. The main molecule has a large hole structure, which is enough to contain the guest molecules and form a molecular capsule.

 

Formation of inclusion complexes: cyclodextrins and their derivatives generally form inclusion complexes through the non covalent combination of guest molecules and cavities. The inclusion reaction is a dynamic process, in which the guest molecules are constantly compounded with cyclodextrin and decomposing at the same time. Hydroxypropyl cyclodextrin is easier to form inclusion complex with suitable guest molecules than original cyclodextrin, because hydroxypropyl replaces 2 and 3 hydroxyl H atoms to increase the length of cyclodextrin cavity, and the hydroxyl on hydroxypropyl may also form new hydrogen bonds with guest molecules to increase the stability of inclusion complex. Most studies show that hydroxypropyl cyclodextrin mainly forms 1:1 inclusion complex with guest molecules, but also forms 2:1 inclusion complex or oligomer.

 

Guest release: part of the purpose of cyclodextrin and its derivatives forming inclusion complexes with guest is to protect, slow release or solubilize. In this case, guest molecules will eventually be released from cyclodextrin. The main factors leading to or accelerating the release of guest molecules are concentration change and competitive substitution, and temperature and pH have little effect. When the solution concentration changes greatly, the guest will release in a very short time. The cyclodextrin compound drug will be released soon after injection into the body, which has almost no effect on the original pharmacology of the drug. When the binding constant of cyclodextrin and guest molecule is 10-4m-1, the decrease of concentration is enough to release the guest. However, for inclusion complexes with high binding power, the effect of concentration gradually decreases or even has no effect, and competitive substitution becomes the main factor. Methanol, ethanol and other small molecules can competitively replace the guest of the inclusion complex. The binding constant is determined by the properties of cyclodextrin and guest molecules. Hydroxypropyl cyclodextrin has higher binding constant than parent cyclodextrin.
